F4为毒性因子，取值1-10。急性毒性下可能使用，如非遗传毒性致癌性、神经毒性或致畸性；F5为可变因子，当没有建立无作用水平时可能使用，当只有LOEL时，根据毒性大小可能使用最大为10的因子。但是NOEL怎么得出，需要毒理学家或者毒理背景的人员，通过综合评判计算得到。
对于有些化学物质，不一定有非常全面的数据，就需要通过实验来提供。
即使是有数据库，面对浩如烟海的文献，怎么选择合适的数据对于药厂或者小型药企来说十分困难，况且药企也没有专业的人员有能力去撰写PDE的报告。
最好的方法就是委托别的机构来写，或者找GMP咨询公司来做。
