三种最常用的培养基是什么?三种最常用的培养?
1.基础培养基(minimum media)在一定条件下含有某类微生物生长繁殖所需的基本营养物质的培养基,也称为基本培养基.
2.加富培养基(enriched media)在普通培养基(如肉汤蛋白胨培养基)中加入某些特殊营养物质制成的一类营养丰富的培养基.用来培养营养要求比较苛刻的异养型微生物.这些特殊营养物质包括血液、血清、酵母浸膏、动植物组织液等.根据待分离微生物的特点设计的培养基,用于从环境中富集和分离某种微生物.(目的微生物在这种培养基中较其他微生物生长速度快,并逐渐富集而占优势,从而容易达到分离该种微生物的目的.)
3.选择性培养基(selected media)一类根据某微生物的特殊营养要求或其对某化学、物理因素的抗性而设计的培养基,具有使混合菌样中的劣势菌变成优势菌的功能,广泛用于菌种筛选等领域.(1)利用该分离对象对某种营养物有一特殊“嗜好”的原理,专门在培养基中加入该营养物,把它制成一种加富性选择培养基(enriched selected media),采用了这类“投其所好”的策略,就可使原先极少量的筛选对象很快在数量上接近或超过原试样中其他占优势的微生物,从而达到了富集或增殖(enrichment)的目的.(2)利用该分离对象对某种抑菌物质所特有的抗性,在筛选培养基中加入这种抑菌物质,经培养后,使原有试样中对此抑制剂表现敏感的优势菌生长大受抑制,原先处于劣势的分离对象大量增殖,最终在数量上占优势.通过这种“取其所抗”的办法也可达到富集培养的目的.这种培养基称为抑制性选择培养基(inhibited selected media).用作抑制它种微生物的选择性抑菌剂有染料(结晶紫等)、抗生素、脱氧胆酸钠和叠氮化钠等.用于选择性的其他理化因素还有温度、氧、pH和渗透压等.
4.鉴别性培养基(differential media)一类在成分中加有能与目的菌的无色代谢产物发生显色反应的指示剂,从而达到只需肉眼辨别颜色就能方便的从近似菌落中找出目的菌菌落的培养基.用于鉴别不同类型微生物的培养基特定的化学反应,产生明显的特征性变化,根据这种特征性变化,可将该种微生物与其他微生物区分开来.最常见的鉴别性培养基是伊红美蓝乳糖培养基,即EMB(Eosin Methylene Blue)培养基.它在饮用水、牛奶的大肠菌群数等细菌学检查和在 E.coli 的遗传学研究工作中有着重要的用途.
三种最常用的培养基是什么?三种最常用的培养?
第一种:胰酪大豆胨琼脂培养基(TSA)
用途:用于设备、车间、人员、包装材料等表面微生物的测定
组成成分:胰酪胨15g;大豆木瓜蛋白酶水解物5g;氯化钠5g;琼脂15g;蒸馏水1000ml;pH7.3±0.2(25℃)
外观:淡黄色、凸起、表面平整、无裂痕、无肉眼可见杂质和气泡。
理化:pH合格,凝胶强度500650g/cm2。
无菌性:20-25℃,培养3天,30-35℃,培养2天,无菌生长,培养基无明显失水现象。
生长试验:接种大肠埃希氏菌、铜绿假单胞菌、枯草芽孢杆菌、金黄色葡萄球菌、白色念珠菌、黑曲霉,生长良好且菌落直径和菌落特征典型。
第二种:胰酪大豆胨液体培养基TSB
TSB培养基用于细菌的增菌、复壮等;配制方法:称取本品30.0g于1L蒸馏水或去离子水中(可按比例增加或减少配制量),微温溶解,分装,121℃高压灭菌15分钟,备用。(中华人民共和国药典2015版培养基) 配方 g/L

第三种:R2A琼脂培养基
R2A琼脂培养基(R2A Agar Medium)用于水中菌落总数的测定;
R2A琼脂培养基配制方法:称取本品18.1g于1L蒸馏水或去离子水中(可按比例增加或减少配制量),加热煮沸至完全溶解,分装,121℃高压灭菌15分钟,灭菌结束后请摇匀,以防琼脂沉积于器皿底部而凝固,备用。
