1 甘草酸的结构及特性
甘草酸是由甘草中提取。甘草，又名美草、蜜甘、甜根子，多生长在我国西北、华北、东北地区，为多年生草本植物。甘草酸作为其主要成分，随产地不同含量亦不同，一般在(4～14)％之内。甘草酸是甘草甜味的有效成分，又名甘草甜素、甘草皂甙。分子式C42H62O16，相对分子质量822．92，其结构式如图1。
甘草酸为白色或淡**结晶型粉末，熔点220 ℃，有特殊甜味，其甜度约为蔗糖的250倍，溶于热水和热的稀乙醇，不溶于无水乙醇和乙醚。甘草酸遇酸则沉淀，常利用此性质进行提取和精制。为了使用方便，一般都把甘草酸制成水溶性的盐类，如甘草酸钠、甘草酸二钠、甘草酸三钠、甘草酸铵等。
2 甘草酸的制备
甘草酸的制备方法有溶剂萃取法，大孔树脂吸附法和植物组织培养法等多种，现介绍采用第一种方法从甘草中提取甘草酸的工艺过程。
2．1 工艺流程
甘草挑选除杂→粉碎→提取→过滤→浓缩→分离→沉淀→重结晶→干燥→成品。
2．2 工艺过程
（1） 粉碎：将干净干燥的甘草放入粉碎机中粉碎，过10目筛选，制得甘草粉。
（2） 提取：称取一定质量的甘草粉放入反应器中，加入其5倍质量的水，在搅拌下于85 ℃以上加热回流2．5 h，过滤、滤渣再加3倍质量的水重复提取一次，合并泸液。
（3） 浓缩：将甘草酸提取液用薄膜蒸发器进行真空浓缩，当滤液体积减少4／5时，趁热过滤。
（4） 分离：在已经冷却的浓缩滤液中，加入其1／2容量的95%的乙醇，然后静止过夜，经过滤除去植物蛋白、多糖等沉淀物。
（5） 沉淀：在经上述处理的甘草酸浓缩液中，用浓硫酸调其PH值，使甘草酸沉淀析出，然后进行离心分离，得粗甘草酸。
（6） 重结晶：用(60～70) ℃的稀乙醇进行重结晶，减压过滤后得甘草酸湿品。
（7） 干燥：将湿甘草酸放入真空干燥箱内，调节真空度，在(70～80) ℃温度下加热(40～60) min，即得干甘草酸。
（8） 成品：将干燥的甘草酸粉碎，过筛，即得甘草酸成品。
3 甘草酸的定性和定量测定
3．1 甘草酸的定性鉴别
取样品4 mg，置白磁板上，加4%硫酸7滴，渐渐变成橙**至橙红色。
3．2 甘草酸纯度的测定
取样品6g，精确称定，加蒸馏水50 mL溶解后，移置100 mL容量瓶中，用乙醇稀释至刻度，摇匀，静置10 h。吸取上清液250 mL置小烧杯中，加氨试液3滴。然后加热至稠膏状，加30 mL蒸馏水溶解，慢慢加入3%盐酸溶液5mL，在冰水中冷却30 min，过滤，沉淀物用5mL冰水洗3次。弃去洗液及滤液，将沉淀物放在表面皿中使水分自然蒸发，然后加入10 mL65 ℃的乙醇，使沉淀物溶解，过滤，将乙醇溶液放入干燥恒重的烧杯中加热蒸干，精确称定。则甘草酸纯度(X)的计算采用下式：

4 甘草酸的应用
甘草酸是甘草甜味的有效成分，通常使用的是其水溶性的盐，称为甘草甜素。它的特点是高甜度，低热能，安全无毒，起泡性和溶血作用很低。因此，在医药、食品、化妆品等行业广泛使用。
甘草酸甜素能与多种生物碱、抗生素、氨基酸等生成复盐或复方制剂。常具有协同、增溶、增加药物稳定性，提高生物利用度及降低毒副作用的功效。甘草酸的许多金属盐，人体可适当吸收，不易造成元素的积蓄中毒，常用来配制健脾开胃、止咳化痰、顺气止喘、治疗慢性肝炎、降低血脂的良药。日本学者对甘草酸甜素的抗病毒性进行了筛选，表明它对艾滋病毒(HIV)的增殖及对细胞变性有抑制作用。1986年日本报道甘草对艾滋病毒的抑制率高达98%，因此，甘草被称为战胜AIDS病的“仙草”。
甘草甜素作为天然的甜味剂与人们的生活密切相关，在食品方面有着广泛的应用。蔗糖虽久负盛名，但糖尿病、肥胖症、高血压、心脏病等患者慎用或禁用。糖精虽低热，但由于是人工合成，加之甜味不纯不受欢迎。而甘草甜素可弥补上述甜素的弊端。甘草酸分子中两分子葡萄糖与配糖体结合，在口腔酶的作用下，经过几秒钟便开始水解出葡萄糖，产生甜味，能持续较常时间，食用后口腔无余酸，集甜味与保健于一体，广泛用于软饮料、罐头、糖果、面制品、酱油、腌制品等方面。
止渴、保健等饮料中加入适量甘草甜素，可以克服多用糖而引起的发酵、酸败等缺点；口香糖、巧克力等糖果中加入甘草甜素，有杀菌、洁齿、消炎、润喉等功效；在酱油中加入甘草甜素，作矫味剂，可抑制盐味和其苦味，增加酱油的风味；在蛋糕、面包、饼干等面制品加入甘草甜素，起到增甜、疏松起泡、柔软化的效果；在腌制品中加入甘草甜素，可克服因少加糖而出现的发酵腐败、变色、硬化等弊端，起到抑腥臭增香甜的作用。
甘草甜素水溶液具有水凝性，可制成变凝性的化妆水剂或乳液，便于使用和保存。由甘草甜素配制的护肤霜、祛斑霜、高级珍珠膏等化妆品，既有美容护肤效果，又有消炎、抗变态反应和治疗皮肤病的作用。
在卷烟方面，甘草甜素作为高级香烟的甜润、增香和保湿剂，可使烟味柔和，减少烟的毒害作用，或许对无害香烟，保健香烟的研制有所裨益。
